Amphotericin B (AmB), a potent but toxic drug, can be effectively utilized in treating fungal infections and leishmaniasis when encapsulated within appropriately structured micelles. This study is centered on designing micelles that contain AmB to reduce toxicity and enhance its antifungal properties. To achieve this goal, AmB was enclosed in micelles based on 1,2-distearoyl-S-glycero-3-phosphorylcholine (DSPC) within a sensitive range of structural alteration, employing different ratios of DSPC and DSPE-PEG2000 (10:1, 9:2, 8:3; w/w) through the solvent evaporation technique in a methanol/chloroform mixture.
DSPC Micelles Preparation and AmB Encapsulation
The micelles were prepared by first ultrasonic treatment and then using the solvent evaporation method with slight modifications. Three different formulations were prepared at 10:1, 9:2, and 8:3 (DSPC: DSPE-PEG2000; w/w) and dissolved in a methanol-chloroform mixture (1:3 v/v ). The mixture is ultrafiltrated to remove solid insoluble components and pyrogens. The filtered solution was transferred to a round-bottomed flask and the solvent was removed using a rotary evaporator at 65 °C under vacuum, leaving a thin layer of lipid components in the flask. Among the three compounds, the drug loading effect of 9:2 w/w was the best. Then, 1 mg of AMB was added simultaneously to the flask containing dry fat as a hydration medium (pH 7.4). Stir the mixture using a magnetic stirrer for 5 minutes and disperse the dry lipids into the hydrated liquid. At this point, microstructures are formed in the form of multilamellar vesicles (MLVs). To reduce the micelle size to nanomicelles, a sonification step was performed using a probe sonifier. Use an ultrasonic homogenizer to ultrasonicate the MLV flask for 3 minutes under certain amplitude and time conditions to form small unilamellar vesicle (SUV) micelles.
